METABOLIK SINDROM VA ABDOMONAL SEMIZLIK BO'LGAN BOLALARDA FTO (RS9939609) VA PPARG 2 (RS18012820) GEN POLIMORFIZMLARI ASSOTSIATSIYASI

Abstrakt

Ushbu  maqolada metobolik sindrom va abdominal semizligi bo’lgan  hamda normal tana massasiga ega bo’lgan bolalarning  FTO (rs9939609) va PPARG 2 (rs18012820) gen polimorfizmi taqsimlanish chastotasining o'rganish natijalari keltirilgan. Tadqiqot natijalari oilaviy shifokorlar, pediatrlar va bolalar endokrinologlari amaliyotida keng qo'llanilishi tavsiya etiladi.


Tadqiqot maqsadi: abdominal semirish kuzatilgan bolalarda FTO (rs9939609) va PPARG 2 (RS18012820) gen polimorfizmlari assotsiatsiyasining xususiyatlarini va MS ko'rinishlarini o'rganish.Materiallar va tadqiqot usullar: Qorin bo'shlig'ida semirib ketgan va metabolik sindrom namoyon bo'lgan 72 bola va normal tana vazniga ega 40 bola tekshirildi.              


Materiallar va usullar. Tadqiqot davomida 72 ta abdominal semizlik hamda metabolik sindrom namoyon bo’lgan va 40 ta normal tana vazniga ega bo’lgan bolalar tekshirildi. Barcha bolalarga antropometrik, anamnestik, klinik, biokimyoviy va molekulyar genetik hamda FTO (rs9939609) va PPARG 2 (rs18012820) gen polimorfizmi taqsimlanish chastotasini aniqlash uchun keng  qamrovli tadqiqotlar o’tkazildi. Olingan ma'lumotlarga statistik ishlov berish Statistica 10 dasturi yordamida shaxsiy kompyuterda amalga oshirildi. Genetik tadqiqotlar davomida allel chastotalari va allel birikmalarining chastotalari hisoblab chiqildi va ularning c2 mezoniga ko'ra Xardy-Vaynberg muvozanatiga mosligi nol gipotezani rad etib, P <0,05 ekanligi aniqlandi.              


Tadqiqot natijalari: Natijalar shuni ko'rsatdiki, abdominalsemizlik va metabolik sindromli bolalarda FTO genining gomozigot genotipi - T/T kamroq(c2 = 4,530, p = 0,034), hamda FTOning gomozigot genotipi A/A gen metabolik sindromli bolalarda sezilarli chastotada tez-tez uchrashi bilan ustunlik qildi. Abdominal semizligi bo’lgan bolalarda nazorat guruhidagi bolalarga (76,2% va 60,0%) nisbatan Pro alleli va Pro12Pro genotipi (85,5% va 73,6%) ustunlik qildi. PPARG-2 genining Pro12Pro genotipi metobolik sindrom namoyon bo’lgan  bolalarda statistik jihatdan ustun ekanligi 84,6% - ushbu sindromga ega bo'lmagan bolalarga nisbatan 62,5% (c2=5,304, p=0,022) ekanligi aniqlandi.


Xulosa: A/A genotipi va FTO genining alleli A (rs9939609) abdominal semizlik va metabolik sindromli bolalarda moyillik bo'ladi. Metobolik sindrom belgilari bilan kechuvchi abdominal semizligi bo’lgan bolalarda Pro12Pro genotipi, metopolik sindrom belgilarisiz kechuvchi abdominal semizligi bo’lgan bolalarda esa 12 Аla alleli uchrash chastotasi metobolik sindrom bo’lgan bolalarga nisbatan statistik jihatdan yuqori bo’ladi, bu ushbu allelni metobolik sindrom rivojlanishida protektivga kiritishga imkon beradi.
